Naproxen, which is similar to ibuprofen in that it inhibits both COX-1 as well as COX-2, can be considered non-selective. The selectivity of drugs in the NSAID group can have a significant impact on side effects and utility.

How Naproxen is Metabolized

Naproxen is absorbed entirely in the GI tract. It usually starts working within 30 minutes to an hour. It may take longer to work if it is taken with food. The peak effect occurs in 2-4 hours, and the half-life lasts 12-17 hours. The half-life is a term used in pharmaceutical terms to refer to the time required to reduce the concentration by 50%. Although the drug is more likely to remain in your body for longer periods of time, its effect usually diminishes significantly after 50 percent. Naproxen is broken down to its primary metabolite, 6-Odesmethyl naproxen. It is then excreted extensively in urine.

How to Take

Naproxen can be purchased over-the-counter at a dose of 220 mg every 8-12 hrs (2-3 times daily), for a maximum daily intake of 660 mg. Adults can take 1500 mg of naproxen per day (naproxen sodium, 1650 mg). For best results, it is recommended that you take the medication with a glass of water. However, it can be taken with food and milk if necessary.

Side Effects

Naproxen can cause nausea, vomiting, heartburn, stomach pain, and constipation in less than 10%. Patients with GI and cardiac conditions should be cautious as they may be at greater risk of their condition getting worse. You should also be cautious when taking blood thinners such as warfarin, Eliquis, Clopidogrel (Plavix), or Heparin.
